      Aims

The aims of MPN&MPNr-EuroNet’s Training School to be held in Nîmes, France, May 11-13 2011 are the following:

      1. to acquire competences in the various techniques designed to detect the JAK2 and MPL mutations characteristic of Philadelphia-negative myeloproliferative neoplasms (MPN). The training school is organized in two sessions:
        • Morning plenary session/lectures: theory of the techniques.
        • Afternoon practical session: applicants will perform the different assays, assisted by experts.
      2. to get the latest information on the different mutations leading to MPN or to MPN transformation (GGCC/46.1 haplotype, TET2, CBP Alpha, CBL, ASXL-1, IDH-1, EZH, LNK…)
      3. to start a cooperative group which could share samples, reference materials and hopefully, generate original studies.

      Objectives

Applicants will acquire theoretical and practical competences enabling them to set up in their own laboratory the assays necessary for the detection and quantification of JAK2V617F, as well as assays for the detection of others mutations in the JAK2 and MPL genes.

The training school will deal with:

      • allelic burden quantification of the JAK2V617F mutation, using real time or LNA clamped PCR
      • HRM (High Resolution Melting analysis) detection of “second line mutations” of JAK2 and MPL: JAK2 exon12 mutations; mutations in other JAK2 exons; MPL exon10 mutations
      • Information and contacts with laboratories expert in the detection of rare mutations (TET2, CBP Alpha, CBL, ASXL-1, IDH-1, EZH, LNK …).

      Keywords

Molecular diagnostic, myeloproliferative neoplasms, JAK2 mutation, MPL mutation, allelic burden quantification
